How To Inject Semaglutide Injection (semaglutide reconstitution)?

The semaglutide STEP trial used the following doses. Here is how to create a solvent mix of the same semaglutide doses.

Using the 2mg semaglutide vial

  • For 0.25 mg semaglutide dose: Add 2 mL bacteriostatic water to vial. Draw 0.25 mL semaglutide solution into syringe (you can do this 8 times)
  • For 0.5 mg semaglutide dose: Add 2 mL bacteriostatic water to vial. Draw 0.5 mL semaglutide solution into syringe (you can do this 4 times)
  • For 1 mg semaglutide dose: Add 1 mL bacteriostatic water to vial. Draw 0.5 mL semaglutide solution into syringe (you can do this 3 times)
  • For 1.7 mg semaglutide dose: Add 1 mL bacteriostatic water to vial. Draw 1 mL semaglutide solution into syringe (you can do this 1 time).

Using the 5mg semaglutide vial

  • For 0.5 mg semaglutide dose: Add 2 mL bacteriostatic water to vial. Draw 0.2 mL semaglutide solution into syringe (you can do this 10 times)
  • For 1 mg semaglutide dose: Add 1 mL bacteriostatic water to vial. Draw 0.2 mL semaglutide solution into syringe (you can do this 5 times)
  • For 1.7 mg semaglutide dose: Add 1 mL bacteriostatic water to vial. Draw 0.33 mL semaglutide solution into syringe (you can do this 3 times)
  • For 2.4 mg semaglutide dose: Add 1 mL bacteriostatic water to vial. Draw 0.5 mL semaglutide solution into syringe (you can do this 2 times)

*This column tells you how much bacteriostatic water you should add to the vial.
**This column tells you how much of the produced solution you should draw up in the syringe so that the syringe contains the desired dose.
***This column tells how many times you can draw up the desired dose with the syringe per vial.

How to Store Reconstituted Semaglutide

Temperature

Semaglutide must be stored in the refrigerator to keep the solution stable. Ideally, the temperature should be within 36°F to 46°F (2°C to 8°C).

Labeling

Label each vial with the concentration of the solution, the date of mixing (reconstitution), and if being used in a laboratory by multiple users, you could add the initials of the person who mixed it (for traceability).

Aliquoting

Think about splitting the reconstituted peptide solution into smaller portions. This approach helps avoid multiple freeze-thaw cycles, which can reduce the likelihood of degradation.

Avoid Freeze-Thaw Cycles

Minimize the number of times you change the temperature of the semaglutide.
